May not be the best, but it's the scene that came to my mind at first.

"Salvation lies within" from "The Shawshank Redemption".

Not the best, but definitely deserving to be among the best.

I love final reveals, and especially 'mysterious title' reveals.

Here is one from an obscure but tense Australian war movie. I've watched it years ago on the tv and could only find a a turkish version.
The movie is about a ruthless sniper duel in the very last days of World War II.
Film tricks the viewer to suspect, that the title adresses it's action time - presumably the last battle of the war...

The reveal shows something else...

Japanese veteran and his brother kill the protagonist's squad, and the protagonist kills the younger Japanese. Then the survivors hunt each other, both seeking revenge. When the opponents finally meet in an open field, each having hardly any ammunition, the Australian is distracted by a little child in a distance walking straight towards a land mine. Instead of fighting his enemy, the hero fires his last ammo at the mine. He misses. The Japanese sniper, recognizing what the Australian has just done, turns in the same direction, and follows up with his last bullet.

just post the whole movie here, for me it is the conversation of landa and that farmer in the first scene that got stuck forever, incredible acting..

From one of my favorite movies:

One of my favourite movie scenes... This one brings a strong confidence when optimus fights on with one hand. Megatron says "Besides, who are you without me... Prime." after beating up Sentinel and Optimus says "Time to decide that!" I like this dialogue very much. And optimus kills sentinel for his betrayal. This scene emotionally touched my heart.
